目的:检测组蛋白去甲基化酶KDM7A对HBV复制的影响。方法:利用实时荧光定量PCR、Western Blot、ELISA检测KDM7A对HBV复制的影响;利用双荧光素酶报告基因检测KDM7A对HBV相关启动子的影响。结果:HBV可以促进KDM7A mRNA的转录及蛋白表达;KDM7A可促进HBV基因组的复制及病毒抗原的产生,增强HBV相关启动子的激活。结论:组蛋白去甲基化酶KDM7A促进HBV的复制。

OBJECTIVE: To detect the effect of histone demethylase KDM7A on HBV replication. METHODS: The effects of KDM7A on HBV replication were detected by real-time fluorescence quantitative PCR, Western Blot and ELISA; the effects of KDM7A on HBV-related promoters were detected using dual luciferase reporter genes. RESULTS: HBV can promote the transcription and protein expression of KDM7A; KDM7A can promote the genome replication and viral antigen production of HBV and enhance the activation of HBV-related promoters. CONCLUSION: Histone demethylase KDM7A promotes the replication of HBV.
