In order to find the most suitable formulation for each growth stage, and establish the rapid propagation system, Pleione formosana embryoes were used to study the effects of different basic culture medium, plant growth regulators and additives in orthogonal experimental design.. Results indicated that: the optimum medium for the embryo germination and later growth was MS + NAA 0.5 mgoL-1 + peptone 2% . The germination rate reached 88.20%, and the plants grew strong. The optimum medium for cultivating strong plant was TH + NAA 0.5 mgoL-1 + 6-BA 1.0 mgoL-1 + peptone 2%. The optimum medium for proliferation of cluster buds and pseudobulb expanding was MS + 6-BA 0.2 mgoL-1 + NAA 1.0 mgoL-1 + mashed potato 100 goL-1 . The average proliferation rate was 4.17,and the diameter of pseudobulb reached 0.33 cm. The optimum medium for rooting was MS + 6-BA 1.0 mgoL-1 + NAA 1.0 mgoL-1 + banana mud 50 goL-1 + mashed potato 50 goL-1. The rooting rate was 78%,and the number of roots was 4.67 per plant.
