原花色素是葡萄果实中的一类重要的多酚代谢产物,对葡萄和葡萄酒的口味有着重要的影响。近年来,一些涉及葡萄果实原花色素生物合成的调节基因得以被发现,其生理功能也得到了阐明。特别是对葡萄MYB家族一系列转录因子的发掘,极大的推动了本领域相关研究的发展。而到目前为止对这些调节基因启动子结构和功能的研究还非常有限。本研究旨在克隆与酿酒葡萄原花色素生物合相关的转录因子VvMYBPA1、VvMYBPA2和VvMYB5a的启动子序列,通过生物信息学对其潜在功能进行预测,通过启动子重组载体侵染烟草叶片来检验其基本功能。研究发现,与原花色素生物合成有关的这些关键结构基因的启动子相比,这些调节基因的启动子序列在潜在功能上有着极大的不同,特别是光响应元件在种类和数量上的差别较大,且多含有其它转录因子和不同激素信号的响应位点。而启动子活性检验则表明,这些目标序列具有启动子的基本活性,且UV-B照射可以导致报告基因表达量的略微增加,表明它们具备一定的响应紫外诱导的功能。

Proanthocyanidins are an important group of polyphenolic metabolites in grape berries, which have great influences on the taste of grape and wine. In recent years, some regulatory genes related to proanthocyanidin biosynthesis in grape berries were found, and their physical functions were also clarified. Especially the studies of a series of regulator of MYB family in grapes have greatly developed the researches in this area. However, until now the researches for the structures and functions of the promoters of these regulatory genes are still quite limited. In the present study, the promoter sequences of the regulatory genes VvMYBPA1、VvMYBPA2 and VvMYB5a which are related to proanthocyanidin biosynthesis were cloned, their potential functions were predicted with the help of bioinformatics, and their basic functions were tested by using infecting of promoter combinant vector on tobacco leaves. This research revealed that compared to the structure genes related to proanthocyanidin biosynthesis, the promoters of these regulatory genes were quite different in the structure and the protentional functions, especially in the kinds and numbers of the photoresponse elements, and they often contain the response sites to other different regulators or phytohormone signals. Besides, the function test revealed that all these aimed sequences had the basic functions of promoters, and after the irraidation of UV-B the expression of the report genes increased slightly, showing their function in responding the induction of UV.
