促血管生成素-1（Angiopoietin-1）对人胃癌细胞粘附和转移作用的研究
Effects of Angiopoietin-1 on attachment and metastasis of human gastric cancer cell line BGC-823
目的：探讨促血管生成素-1(Angiopoietin-1，Ang-1)对人胃癌细胞中整合素β1（integrin β1）和CD44V6的影响，初步研究其对肿瘤粘附和转移的影响及可能的作用机制。方法：利用腺病毒作为载体将已构建成功的含Ang-1基因的重组腺病毒（Ad-Ang-1）瞬时转染人胃癌细胞株BGC-823，转染Ad-GFP作为阴性对照，未转染的正常细胞作为空白对照，分别使用半定量RT-PCR和Western blot方法对三组细胞中整合素β1和CD44V6的mRNA和蛋白表达水平进行分析；应用免疫细胞化学法检测转染前后整合素β1和CD44V6的蛋白表达；细胞粘附法检测转染前后细胞与细胞外基质（ECM）粘附能力的改变。结果：RT-PCR、Western blot结果显示整合素β1和CD44V6 mRNA在Ad-Ang-1组中的表达要明显高于空白对照组及Ad-GFP组(P<0.05)；免疫细胞化学法也显示Ad-Ang-1组的整合素β1和CD44V6表达强度高于空白对照组和Ad-GFP组(P<0.05)；细胞粘附实验显示转染Ad-Ang-1后，细胞与细胞外基底之间的粘附力明显增强(P<0.05)。结论：转染Ang-1能够明显提高人胃癌细胞BGC-823中整合素β1和CD44V6 mRNA、蛋白的表达，从而促进肿瘤细胞的粘附和转移。
Objective: to explore the effects of Angiopoietin-1 on integrin β1 and CD44V6 of human gastric cancer cell line BGC-823 and study the mechanism of adhesion and metastasis on gastric cancer. Methods: The cell line of BGC-823 was transfected with Ad-Ang-1 and Ad-GFP. The expression of integrin β1 and CD44V6 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The protein expression of integrin β1 and CD44V6 was detected by Western-blot. The expression of integrin β1 and CD44V6 was measured by immunohistochemistry. The method of cell adhesion can present the relationship between cell and ECM. Results: Ang-1 transfected BGC-823 successfully. RT-PCR and Western-blot revealed that the mRNA and protein expression of integrin β1 and CD44V6 increased in Ad-Ang-1transfected group (P<0.05). Compared with mock transfected group and Ad-GFP group, integrin β1 and CD44V6 expression intensity greatly increased (P<0.05). Adhesion assay also presented an increase. Conclusion: Transfection of Ang-1 into human gastric cancer cell line BGC-823 can significantly promote expression of integrinβ1 and CD44V6, and increase in adhesion and metastasis to ECM.
