目的 建立和优化鸡毒支原体蛋白组学研究的双向电泳方法。 方法 以改进的超声辅助的尿素裂解法为总蛋白提取方法,以固相pH梯度等电聚焦为第一相和垂直SDS-PAGE为第二向,对样品制备、上样量、水化时间、等电聚焦程序、SDS-PAGE电泳程序及染色方法等相关技术进行了比较分析和优化。 结果及结论 成功得到了鸡毒支原体的双向电泳图谱,图谱分辨率高、背景清晰、蛋白点数多、重复性好,为进一步进行鸡毒支原体的比较蛋白组学研究奠定基础。

Objective To establish and optimize two-dimensional gel electrophoresis method for proteomic analysis of Mycoplasma gallisepticum. Methods Improved ultrasound-assisted urea lysis method is used as extraction method for the M. galliseoticum proteome. Immobiline pH gradients isoelectric focusing was used as 1D and vertical SDS-PAGE was used as 2D. Some factors, such as protein extraction method, volume of loading sample, hydration time, isoelectric focusing procedure, SDS-PAGE electrophoresis procedure, protocol for staining the gels, were improved. Results and Conclusion Two-dimensional gel electrophoresis method of M. gallisepticum was developed, providing a clear background, high resolution, large number of protein spots, and good reproducibility. Optimized 2-DE method can be effectively applied in future research of comparative proteomics study of M. gallisepticum.
