四氢生物蝶呤合成抑制剂对小鼠骨髓基质细胞增殖分化的影响
Effect of BH4 synthesis inhibitors on the proliferation and differentiation of op9 cells
GTP 环化水解酶 (GTPCH) 是四氢生物蝶呤 (BH4) 合成途径中的限速酶,2,4-二氨基-6-羟基嘧啶(DAHP)是GTPCH的抑制剂。本文采用四甲基偶氮唑盐(methyl thiazolyl tetrazolium, MTT)法,鸡尾酒诱导分化法和油红O染色法,检测DAHP对 op9小鼠骨髓基质细胞增殖和分化的影响,探讨BH4与脂质合成的关系。MTT结果显示,在一定浓度范围内DAHP呈剂量依赖关系的抑制op9细胞增殖。倒置显微镜观察和油红染色检测结果显示,在500 μmol/L至900 μmol/L之间DAHP对op9细胞分化为脂肪细胞具有明显抑制作用,这表明BH4可能在脂质合成中有重要作用。荧光定量PCR反应分析表明,DAHP使GTPCH转录水平发生下调,表明DAHP成功抑制了BH4合成途径。同时,DAHP还会抑制op9分化为脂肪细胞过程中NADPH的产生,使糖酵解途径、磷酸戊糖途径、TCA循环和叶酸代谢中的产NADPH基因转录水平均发生下调,这可能是BH4影响脂质合成的机制。
BH4 belongs to the aromatic amino acid hydroxylase coenzyme, and is an important cofactor for nitric oxide synthase. GTP cyclohydrolase I (GTPCH) is the rate-limiting enzyme of BH4 synthesis. DAHP is the inhibitor of GTPCH. The effect and mechanism of DAHP on proliferation and differentiation of op9 cells were explored by methyl thiazolyl tetrazolium (MTT) method. Oil red O staining and spectrophotography were applied to analyze the degree of differentiation. DAHP inhibited proliferation of op9 cells in a dose-dependent manner. Oil red O staining results showed that DAHP inhibited differentiation of op9 cells iEffect of BH4 synthesis inhibitors on the proliferation and differentiation of op9 cellsn the range of 500 μmol/L to 900 μmol/L. Real-time PCR was applied to detect the mRNA expression of related genes. The transcription level of GTPCH was decreased. Glycolysis, pentose phosphate pathway, phenylalanine metabolism, TCA cycle, and one carbon pool by folate were down-regulated through NADPH requlation, and all of which may explain why the differentiation of op9 cells was inhibited by DAHP. ?????
