一种新型独脚金内酯受体激动剂的筛选与功能验证
Screen and functional verification of a new type of strigolactone receptor
独脚金内酯SL是一类新型植物激素,在植物株型构成中等发挥关键功能。外源添加SL及其类似物(如GR24)对于农业生产具有重要作用。但是,目前SLs人工合成类似物依然存在合成难度大、稳定性差及成本高的问题。本文基于水稻SL受体OsD14与人工合成类似物GR24复合体的三维结构,以稳定的芳养键代替不稳定的烯醇醚键,构造新的SL类似物结构-gx系列化合物。然后,利用下胚轴抑制实验和酵母双杂交实验,对gx系列化合物的生物活性进行评估,发现gx01、gx03、gx04均能抑制下胚轴的伸长、抑制拟南芥莲座叶分枝、及促进根毛伸长,其中gx04的活性最高,且其活性优于GR24。紧接着,通过HPLC对这些化合物进行稳定性评估,结果表明,gx系列化合物存在活性与稳定性优于GR24。本文结果对进一步开发新型、高效SL类似物提供了重要的理论指导。
Strigolactone(SL) is a new kind of plant hormone, which plays a key role in plant type constitution. Exogenous addition of SL and its analogues (such as GR24) plays an important role in agricultural production. However, there are still some problems in the synthesis of SLS analogues, such as difficult synthesis, poor stability and high cost. Based on the three-dimensional structure of the complex of rice SL receptor OSD14 and synthetic analogue GR24, a new SL analoguegx-series was constructed by replacing the unstable enol ether bond with a stable aromatic bond. Then, the biological activities of gx-series compounds were evaluated by hypocotyl inhibition test and yeast two-hybrid test, the results showed that gx01, gx03 and gx04 could inhibit hypocotyl elongation, inhibit rosette leaf branching and promote root hair elongation, and the stability of these compounds was evaluated by HPLC. The results showed that the gx-series compounds were better than GR24 in activity and stability. The results of this paper provide important theoretical guidance for the further development of new and efficient SL analogues.
