目的:观察"双固一通"电针法对胰岛素抵抗模型大鼠下丘脑中的PI3K及p-PI3K蛋白表达影响。方法:以高脂饮食诱导的诱导胰岛素抵抗模型大鼠。正常组(Group 1)大鼠不给予任何处理,常规饲养;预防组(Group 3)、"双固一通"电针组(Group 4)、脑室灌注脑脊液组(Group 5),和脑室灌注阻滞剂组(Group 6)分别采用"双固一通"法电针治疗。实验开始后的第8w和第16w检测各组大鼠体重(BW)、空腹血糖(FPG)、血浆胰岛素(Fins)水平,及胰岛素抵抗指数(ISI);West-blot 法检测大鼠下丘脑PI3Kp110及p- PI3Kp110蛋白表达。结果: (1) Group 3 与Group 4大鼠BW、FPG、FINS降低(p<0.05),IAI升高(p<0.05); (2) Group 3、 Group 4、大鼠下丘脑中 PI3Kp110蛋白及p- PI3Kp110蛋白表达明显提高(P<0.05)。结论:"双固一通"电针法可通过调节大鼠下丘脑中 PI3Kp110蛋白及p- PI3Kp110蛋白表达,从而改善胰岛素抵抗模型大鼠的体重、空腹血糖、血浆胰岛素水平,胰岛素敏感性。

Objective: To observe the effects of Double-reinforcing and One-unblocking of acupuncture method on on expression of PI3K and p-PI3K insulin resistance model rat. Methods: Use the high-fat diet induced insulin resistance model of rats. Group 1 rats were feed normal food, Group 3, Group 4, Group 5, Group 6 were feed with high-fat diet daily, and treated with Double-reinforcing and One-unblocking of acupuncture stimulation. BW, FPG, Fins, and IAI were measureed in 8th week and the 16th week respectively; the hypothalamus PI3K expression and p-PI3K expression were detected by western-blot.Result: (1) BW, FPG, Fins were decreased(p<0.05), and IAI increased(p<0.05) in Group 3 and Group 4; (2) the hypothalamus PI3K expression and p-PI3K expression were increased in Group 3 and Group 4(P<0.05). Conclusion:The above results suggest that the effects of Double-reinforcing and One-unblocking of acupuncture method can improve the BW, FPG, Fins, and IAI,probable through regulate the hypothalamus PI3K expression and p-PI3K expression in insulin resistance model rat.
