Expression and clinical significance of PTENp1,PTEN and miR-21 in oral squamous cell carcinomas

目的:评价PTEN、PTENp1和miR-21三者在口腔鳞状细胞癌(OSCC)中的临床意义。方法:搜集1992年1月至2007年1月期间62例OSCC组织标本,采用qRT-PCR检测PTEN、PTENp1和miR-21在OSCC组织中的表达情况及其相关性。结果:qRT-PCR检测结果显示PTEN和PTENp1在OSCC组织中的表达显著低于正常组织(P<0.05);而miR-21的在OSCC组织中的表达要显著高于正常组织(P<0.05)。Pearson相关性分析显示:在口腔鳞状细胞癌组织标本中PTEN与miR-21之间无明显负相关关系(R = 0.032,P = 0.6603),而PTEN与PTENp1之间存在显著的正相关关系(R = 0.1970,P = 0.007)。结论:PTENp1可能作为PTEN的ceRNA竞争性调节miR-21的功能,从而调控OSCC细胞的生物学行为。

Purpose:To evaluate the clinical significance of PTENp1,PTEN and miR-21 in oral squamous cell carcinomas. Methods: The 44 cases tissue specimens and paracancerous tissues were collected during January 1992 to January 2007, qRT-PCR was used to detect the expression levels and correlation of PTENp1,PTEN and miR-21 expression in those specimens. Results: qRT-PCR showed that the levels of PTEN and PTENp1 expression were significantly decreased in OSCC tissues compared with normal tissues (P<0.05), while the expression of miR-21 was upregulated in OSCC tissues compared with normal tissues (P<0.05). Furthermore, Pearson analysis demonstrated that no siginificant correlation between PTEN and miR-21 was observed in OSCC tissues(R = 0.032,P = 0.6603), while the expression of PTEN positively correlated with the expression of PTENp1(R = 0.1970,P = 0.007). Conclusion: Our ?ndings suggest that PTENp1, as the ceRNA of PTEN, competitively bind the miR-21 to regulate the biological behaviour of OSCC.
