lthough numerous evidences show that Tumor Necrosis Factor Receptor-Associated Factor 4 (TRAF4) plays an important role in many tumors, the exact molecular mechanism remains elusive. In the present study, we analyzed the expression of TRAF4 with immunofluorescence and western blot. We found that the expression of TRAF4 in normal breast cells MCF-10A was lower than that in breast cancer cells MCF-7 and MDA-MB-231. TRAF4 localized in both cytoplasm and nuclei in MCF-7 and MDA-MB-231. After TNF-α treatment, TRAF4 depletion by siRNA in MCF-7 cells could suppress cell proliferation and the nuclear expression of NF-κB significantly reduced. The percentage of early apoptotic cells in MCF-7 cells was augmented upon TRAF4 knockdown and more G1 phase cells and less S phase cells were detected. These results suggest that TRAF4 has anti-apoptotic effects on apoptosis induced by TNF-α in MCF-7 cells.
