齐墩果酸对阿尔茨海默病模型大鼠海马区ADAM10、sAPPα及凋亡相关蛋白bax和bcl-2表达的影响
Influence of oleanolic acid on the expression of sAPPα, α-secretase protein, and Bax / Bcl-2 in hippocampus of AD rats
目的:观察齐墩果酸对阿尔茨海默病模型大鼠海马区α分泌酶(ADAM10)、可溶性淀粉样前体蛋白(sAPPα),以及凋亡相关蛋白bax、bal-2表达的影响,初步探讨齐墩果酸治疗AD可能性作用机制。方法:将30只雄性SD大鼠随机分为假手术组,模型组和齐墩果酸组,采用右侧侧脑室注射聚集态β-淀粉样蛋白片段25-35(Aβ25-35)片段,造模成功后齐墩果酸组按照20mgoKg-1予以齐墩果酸灌胃,假手术组和模型组予同体积蒸馏水灌胃,灌胃1次/日,连续4周。采用Western bolt检测大鼠海马区ADAM10、sAPPα、bax、bcl-2蛋白的表达。结果:与假手术组相比,模型组海马区ADAM10、sAPPα、bcl-2蛋白表达显著下降,差异具有统计学意义(P<0.05);bax表达量显著升高,差异具有统计学意义(P<0.05);与模型组相比,齐墩果酸组大鼠海马区ADAM10、sAPPα、bcl-2蛋白表达显著增加,差异具有统计学意义(P<0.05);bax表达显著下降,差异具有统计学意义(P<0.05)。结论:补肾中药女贞子有效成分齐墩果酸可通过促进AD模型大鼠海马区ADAM10、sAPPα、bcl-2蛋白的表达,抑制bax蛋白的表达进而达到神经保护的作用。
Objective:To observe the effect of oleanolic acid (OA ) on α-secretase (ADAM10), soluble amyloid precursor protein (sAPPα), and apoptosis-related protein bax, bcl-2 in hippocampus of rat models of Alzheimer's disease, to preliminary explore the possible mechanism of oleanolic acid treating AD. Methods: 30 male SD rats were randomly divided into sham group, model group and oleanolic acid group, and aggregation β- amyloid fragment 25-35 (Aβ25-35) was injected into the right lateral ventricle. After successfully build the AD model, feeding the rats in OA group with oleanolic acid in accordance with 20mg o Kg-1, and rats in sham group and model group were given the same volume of distilled water, once / day, for 4 weeks. Western blot was applied to detect ADAM10, sAPPα, bax, and bcl2. Results: Compared with sham group, ADAM10, sAPPα, bcl-2 expression in model group hippocampus was significantly decreased, and the difference was statistically significant (P <0.05); with significantly increased bax expression, and the difference was statistically significant (P <0.05); compared with the model group, ADAM10, sAPPα, bcl-2 protein expression was significantly increased in oleanolic acid group, and the difference was statistically significant (P <0.05); the expression of bax decreased significantly, and the difference was statistically significance (P <0.05). Conclusions: The ingredient in Fruits of Ligustrum lucidum, an herb for tonifying kidney, oleanolic acid could promote the expression of ADAM10, sAPPα, bcl-2, and inhibit the expression of bax in hippocampus of AD model rats to achieve a neuroprotective effect.?????
