本研究从'王族'海棠中获得了编码bHLH转录因子的全长cDNA序列,命名为MRYLC。生物信息学分析结果表明,该序列编码区域共1956bp,编码651个氨基酸,其蛋白质相对分子质量为72.9kDa,理论等电点为5.51,位于细胞核的可能性最大。采用实时荧光定量研究了不同发育时期的MRYLC基因的表达水平,利用高效液相色谱分析色素变化情况,结果显示,随着叶片发育,MRYLC基因的表达量逐渐降低,与叶片中呈色色素的含量变化相关。
he full-length cDNA of abasic helix-loop-helix (bHLH) gene,which was named as MRYLC,was isolated from Malus'Royalty'. Bioinformatics analysis indicated that the MRYLC gene contains 1956-base pair(bp) open reading frame (ORF) and encodes a 651-amino-acid polypeptide with a predicted molecular weight of 72.9kDa and pI of 5.51,,has the most possibility located in cell nucleus. Real-time PCR analysis indicated that with leaves development, MRYLC expression level gradually reduced, with associated flavonoids changing in leaves ,which indicated by HPLC. .
