NIK is necessary for NF-κB activation by TNFα
在细胞中,一些泛素化修饰的蛋白在调节NF-κB信号通路中起重要作用。为了研究TNIK(Traf2 and NCK-interacting kinase)对于NF-κB信号通路是否是必需的,首先通过western blot 及免疫共沉淀分析,发现加入TNFα处理,TNIK出现泛素化及蛋白酶体调节的下调。通过RNA干扰的进一步研究、western bolt及报告基因活性分析可以发现TNIK对由TNFα引起的NF-κB活化是必需的。故而得出结论:TNIK在TNFα诱导的NF-κB活化的过程中是必需的。
In the cell, some proteins modified by ubiquitin play central role in regulating the NF-κB signaling pathway. The purpose is to study whether TNIK is necessary in NF-κB signaling pathway. By combining western blot and coimmunoprecipitation assays, we found that TNIK underwent ubiquitin- and proteasome-mediated degradation in response to TNFα. Further investigation through RNA interference, western blot and reporter activity assay disclosed that TNIK was necessary for NF-κB activation by TNFα. In conclusion, the finding indicated TNIK was necessary for NF-κB activation by TNFα.
