Fine mapping of the adult plant resistance to powdery mildew locus QPm.caas-4DL in wheat

小麦白粉病是世界范围内影响小麦高产、稳产的重要病害,培育抗病品种是防治小麦病害最为经济、安全有效的措施,而成株抗病基因的合理利用及其有效的分子标记对培育持久抗性小麦品种尤为重要。本实验选用仅含有QPm.caas-4DL的DH系与感病亲本京双16杂交并回交3次后获得598个BC3F2家系的次级群体,通过对该次级群体进行两年白粉病表型鉴定和相关分子标记检测,从而对白粉病成株抗性4DL位点进行精细定位。结果表明:次级群体中,携带成株抗性位点QPm.caas-4DL单株与未携带该位点单株数目比例符合1对等位基因的遗传分离比例,并且获得与该位点连锁距离在1cM之内的SSR分子标记,Xgwm192. 该标记可以有效地应用于遗传群体分析,但育种材料检测需要更为紧密连锁/功能性分子标记。

Powdery mildew is the devastating disease in common wheat (Triticum aestivum L.) worldwide. Deployment of resistant cultivars is the most environmentally friendly and economical way for controlling the diseases, especially for cultivars with adult-plant resistance (APR). In the present study, we selected one double haploid (DH) line carrying QPm.caas-4DL to cross with the susceptible line Jingshuang 16 and backcrossed three times to get 598 lines in BC3F2 generation. This population has used to fine map the APR to powdery mildew in chromosome 4DL. The result showed that there was only one resistance gene in the BC3F2, viz. QPm.caas-4DL and detected one closer linked molecular, Xgwm192, within 1cM away from the resistance locus. This SSR molecular marker can be used in the genetic population, However, it is very necessary to identify much closer or gene-based molecular for wheat breeding.
