家蚕新基因BmY14的表达分析和亚细胞定位研究
Expression Analysis and Subcelluar Localization of New Gene BmY14 from silkworm pupa, Bombyx mori
RNA监视,也称无意义密码子介导的mRNA的降解(nonsense mediated mRNA decay, NMD) 。Y14是一类在真核RNA监视中起主要作用的RNA结合蛋白。我们从家蚕蛹期cDNA文库中获得一条序列长度为501 bp的cDNA,其编码的166个氨基酸组成的蛋白质与已知的RNA结合蛋白Y14序列同源性较高,含有一个保守的RNA结合结构域,我们将它命名为家蚕类Y14基因BmY14。该蛋白质预测分子量为18.4 kDa。我们将其ORF序列克隆到原核表达载体中,构建重组质粒,经双酶切鉴定及序列测定正确后转化大肠杆菌BL21(DE3),用IPTG诱导成功表达该蛋白,并凭质谱鉴定确定表达蛋白为目的蛋白。表达的融合蛋白经亲和层析纯化后,免疫兔获得特异性强的多克隆抗体。利用该抗体以ELISA和Western blot检测家蚕七个发育阶段和五龄幼虫多种组织或器官中BmY14蛋白的表达和分布,发现该蛋白在四龄、五龄幼虫、蛹、蛾四个发育阶段均有表达,其中蛹中表达量最高;各个组织或器官中,气囊中表达量最高,其次是血液和生殖器。利用抗体的免疫荧光标记对家蚕Bm5细胞定位,观察到荧光集中分布在细胞核,证实BmY14主要存在核中,在家蚕变态发育过程中,特别在生殖器和气囊的成熟过程中可能起重要作用。
Y14 is a kind of RNA-binding protein which plays a major role in RNA surveillance, another name for NMD (nonsense mediated mRNA decay). According to the large-scale scanning of the cDNA library of pupae phase of silkworm, a 501 bp cDNA was identified which encoded a protein had high sequence homology with a known protein Y14, possessing a conservative RNA binding motif. We named this gene BmY14 (Bombyx mori Y14). This gene has an ORF encoding 166 amino acid residues with the predicted molecular weight of 18.4 kDa. The ORF of the gene was cloned into the prokaryotic expression vector and the recombinant plasmid was transformed into E.coli BL21(DE3). After being induced with IPTG, this gene was successfully expressed. Mass spectrogram was carried out to make sure that it was the interested protein. After being purified with Magnet HisTM Ni-Particles, the recombinant protein was immunized as an antigen against a rabbit. ELISA and Western blot analysis revealed that the antibody specificity is high. It was also detected with ELISA and Western blot that the protein was expressed in several silkworm tissues and 4 developmental phases, with the highest expression in pupa in periods, and BmY14 express most in aerocyst, then blood and organs of generation in tissues. Using fluorescent antibody for subcellular localization associated with DAPI counterstain in Bm5 cell, it was found that BmY14 was preliminarily localized in the nucleus. It was confirmed that BmY14 existed mainly in nuclear, and it was proposed that it might play an important role in the silkworm metamorphosis development, particularly, in the maturity of genital and aerocyst.
