低温对两种紫草科植物△6-脂肪酸脱氢酶基因表达的影响
Low temperature regulate △6-fatty acid desaturases at a transcriptional level in Arnebia euchroma and Nonea capsica(willd.) G. Don
【目的】研究两种紫草科植物新疆软紫草(Arnebia euchroma)和假狼紫草(Nonea capsica(willd.) G. Don)中γ-亚麻酸合成关键酶基因△6-脂肪酸脱氢酶基因的表达特性。【方法】以翻译延伸因子EF1a为内参基因,利用半定量逆转录聚合酶链式反应(semi-QRT-PCR)技术研究△6-脂肪酸脱氢酶基因的表达特性;用气相色谱技术分析产物γ-亚麻酸在低温下的积累情况。【结论】△6-脂肪酸脱氢酶基因在紫草植物的叶,茎,根中均表达,在低温诱导不同时间后紫草植物叶片中△6-脂肪酸脱氢酶基因的表达有所增强;产物γ-亚麻酸在低温下积累,这表明紫草科植物叶片中△6-脂肪酸脱氢酶基因表达量的增加可能与适应低温胁迫的逆境有关。
expression analysis of the △6-fatty acid desaturases gene, was carried out by Semi-QRT-PCR,using EF1a gene as reference gene, Semi-QRT-PCR results show that △6-fatty acid desaturases gene was expressed in leaves, stem and root of Arnebia euchroma and Nonea capsica (willd.) G. Don, The transcribing of △6-fatty acid desaturases gene in leaves were slightly enhanced by Low temperature.And the product γ-linolenic acid(GLA)accumulated in the leave under low temperature by gas chromatography analysis.These suggested that expression changed of △6-fatty acid desaturases may be related to helping the plants adapt to low temperature.
