NT(AINTEGUMENTA)基因家族属于植物AP2/ERF这个庞大的转录因子家族中的AP2亚家族,广泛参与植物器官发育调控和生长素信号转导相关途径并发挥重要作用。为了揭示番茄ANT基因家族的功能,本研究利用全基因组信息鉴定番茄ANT转录因子家族,分析其结构特征、系统发育关系、染色体定位及组织表达模式。结果表明:番茄基因组中共有5个ANT基因,分别位于5条染色体上。每个ANT成员都包含2个AP2保守结构域,具备4个特异的motifs和独特的linker region。系统发育关系显示番茄ANT转录因子和拟南芥同源蛋白属于AP2/ERF家族的euANT进化分支,这表明它们可能存在相似的起源方式。组织表达模式分析发现,ANT基因在番茄各个组织中均有表达,在幼嫩的生长器官中表达量较高于成熟器官。ANT基因在番茄茎、叶中微弱表达;在生殖器官花中强烈表达,表达丰度从花蕾到盛花时期呈递增趋势;在果实发育各时期中,幼果时期表达量较高,在绿熟、转色果、黄果及红果时期的表达量相对较低。

NT(AINTEGUMENTA) gene family is a member of the AP2 subfamily witch is in the AP2/ERF family of transcription factors in plant. ANT gene family involves extensively in regulation of plant organs development and playing an important role in Auxin related signaling pathways. In order to study potential function of ANT gene family members in tomato, we investigated ANT family genes with the whole genome database using bioinformatics method. We did analysis of the structure characteristics, phylogenetic relationships, chromosomal location, and tissue expression pattern. The results showed that total 5 members were identified and they distributed in 5 chromosomes of tomato. They all included 2 AP2 conservative structure domain, 4 specific motifs and unique linker-region. Phylogenetic analysis indicated that tomato ANT transcription factors and the homologous gene in arabidopsis belonged to the euANT branch family of AP2 / ERF family, which suggested that they may have a similar origin. Quantitative PCR analysis indicated that all of the ANT genes showed expression in multiple plant tissues, with higher levels of expression in young tissues as compared with older tissues. ANT genes were low in stem and leaf tissues; mainly expressed in flower organ (Bud and flower); detected in immature fruit while very low in mature green, break, yellow fruit and red fruit stage.
