通过体内、外实验探讨骨髓间充质干细胞(bone mesenchymal stromal cells, BMSCs)治疗实验性自身免疫性重症肌无力(Experimental Autoimmune Myasthenia Gravis ,EAMG)的效果和相关机制。 全骨髓培养法提取、扩增健康Lewis大鼠的BMSCs;特异性AChR多肽片段尾根部免疫Lewis大鼠,建立EAMG动物模型;在二次免疫的同时以同种异体BMSCs107/只尾静脉移植,依据临床评分标准,ELISA和流式细胞仪等方法观察和评定BMSCs对EAMG的治疗效果;通过体外BMSCs与AChR特异性T细胞共培养实验,以1-MT封闭BMSCs分泌的吲哚胺二三双加氧酶(indoleamine 2, 3-dioxygenase,IDO),采用ELISA,流式细胞仪法等方法探讨BMSCs治疗EAMG的具体机制。体内BMSCs回输使得EAMG症状明显缓解,各项检测指标均有明显差异,治疗有效;体外实验结果显示,BMSCs能够影响Th细胞相关因子的分泌,改变Th细胞亚群的分布,并且当以1-MT封闭IDO后,这种调节作用在一定程度上被抑制。BMSCs能通过分泌IDO在一定程度上调节四种Th亚群的平衡,进而达到对EAMG的治疗效果。

he purpose of this paper is to investigate the therapeutic efficacy and mechanisms of bone mesenchymal stromal cells (BMSCs) on experimental autoimmune myasthenia gravis (EAMG) both in vivo and in vitro. BMSCs were separated from normal young Lewis rats, and EAMG-model in Lewis rats were established by immunizing R-AChR at the tail root. Intravenous administration of syngenic BMSCs (107/rat) was given to EAMG-model rats on the day of their second immunization. In vitro, AChR-specific T cells were co-cultured with BMSCs at proper ratio, in the presence or absence of 1-MT, the inhibitor of indoleamine 2, 3-dioxygenase (IDO). By the evaluation of clinical score, ELISA and flow cytometry method, the therapeutic effects and the machenisms of BMSCs on EAMG were investigated and found that intravenous administration of syngenic BMSCs effectively ameliorated the pathological features of the disease, and both in vivo and in vitro, the given of BMSCs regulated the imbalance among the Th1, Th2, Th17, and Treg cell subsets, which was at least partially abolished by their secretion of IDO.
