高强度聚焦超声对前列腺癌小鼠的疗效及T细胞亚群的影响

目的 探讨高强度聚焦超声(HIFU)对前列腺癌荷瘤小鼠的疗效及T细胞亚群的影响。 方法 RM-1细胞接种于C57BL6小鼠建立小鼠前列腺癌皮下移植瘤模型,随机分为肿瘤对照组、HIFU组、放疗组和HIFU联合放疗组。观察治疗后各组的肿瘤生长曲线,计算肿瘤体积倍增时间(DT)。2周后,通过流式细胞仪检测正常小鼠和荷瘤小鼠外周血CD3+、CD3+CD4+、CD3+CD8+ 细胞占白细胞总数的百分比。 结果 与肿瘤对照组相比,HIFU组、放疗组肿瘤生长延缓,DT显著延长。HIFU组小鼠的 CD3+(%)、CD4+ (%)显著高于肿瘤对照组,而放疗组与肿瘤对照组无显著差异。HIFU联合放疗组的DT较HIFU组显著延长,小鼠的CD3+(%)、CD4+ (%)显著高于HIFU组,且CD4+ (%)显著高于正常小鼠。 结论 HIFU治疗前列腺癌不仅能局部抑制肿瘤生长,而且能通过对T细胞亚群的影响提高机体的细胞免疫水平。HIFU联合放疗能进一步提高疗效。
Objective To explore therapeutic effects of high intensity focused ultrasound on mice with prostate cancer and its influence on systemic T cell subsets. Methods To establish subcutaneous murine prostate carcinoma model by vaccinating RM-1 cells to C57BL6 mice, mice were randomly divided into control group, radiotherapy group, HIFU group, HIFU combined with radiotherapy group. We observed the growing curve of tumor after treatment and calculated the tumor doubling time (DT). Two weeks later, normal and tumor-bearing mice were observed CD3+, CD3+CD4+ and CD3+CD8+ cells' percentage within the total white blood cell population by flow cytometric analysis. Results Comparing with control group, both HIFU and radiotherapy slow tumor's growth and significantly prolonged the DT. CD3+(%) and CD4+(%) of mice in HIFU group were higher than those in control group, and there was no significant difference between radiotherapy group and control group. The DT of HIFU combined with radiotherapy group was significantly prolonged than that of HIFU group, and its CD3+(%) and CD4+(%) were significantly higher than those of HIFU group, CD4+(%) were significantly higher than those of normal mice. Conclusion HIFU in treatment of prostate cancer not only inhibited local tumor growth, but also improved systemic cellular immunity by influencing T cell subsets. HIFU in combined with radiotherapy further improved its therapeutic effects.
