材料表面成分对干细胞成骨分化的影响的研究

生物材料表面性质的微小差异对细胞的功能有着重要的影响。微图案化技术可以在同一表面上制备具有不同性质的规则分布的微小区域。本文将利用微图案化技术在细胞培养板表面制备具有三种不同化学成分的规则分布的微小区域,并在同一培养环境条件下考察材料表面的化学成分对人骨髓来源的间充质干细胞(hMSCs)成骨分化的影响。本文首先将光反应性基团引入到高分子材料的侧链上,并通过紫外交联仪在细胞培养板表面制备微图案,微图案的完整性和高度分别通过光学显微镜和扫描探针显微镜(SPM)来测定。将hMSCs培养于微图案化材料的表面,并进行成骨诱导分化,通过细胞形态观察、ALP染色、Alizarin Red S染色来检测材料表面化学成分对hMSCs成骨分化的影响。实验结果表明,紫外交联法可以在有机材料表面制备形状规则的微图案,可以在同一表面上制备具有三种不同化学成分的微小区域;微图案化材料表面成分对hMSCs分化有着重要影响,明胶微图案化的微小区域有利于无机物质的沉积。
he slight difference in biomaterial surfaces has important effect on cells functions. Micropatterning technology make it possible to prepare a surface with different surface properties in regular areas. In this study, micropatterning technology was applied to prepare micropatterns with three kinds of different surface properties on cells culture plate surfaces, and the effect of materials surface physicochemical properties on the osteogenic differentiation of hMSCs was investigated in the same environment. In this study, the photo-reactive polymers were synthesized by the introduction of azidophenyl groups into the side chains of polymers, and the micropatterns were prepared by UV-irradiation of photo-reactive polymers on cells culture plate surface, the height and integrity of immobilized photo-reactive polymers were detected by the optical microscope and scanning probe microscope. hMSCs were cultured on the micropattern surfaces, and performed osteogenic differentiation, ALP staining and Alizarin Red S staining were conducted to detect the effect of surface component on the osteogenic differentiation of hMSCs. The results indicated that micropattern with different surface properties in regular morphology can be prepared by UV-irradiation method, and surface component of micropatterns had important effect on the osteogenic differentiation of hMSCs, gelatin micropatterned areas were beneficial for the deposition of minerals during osteogenic differentiation.
