本文采用RT-PCR技术分析了果实成熟过程中两个品种可食部位蔗糖代谢相关酶的表达状况,同时测定了对应时期它们的糖组分含量变化,结果表明在果实成熟过程中酸性转化酶(AI)基因在奉节72-1脐橙和奉晚脐橙间出现表达差异,而蔗糖合成酶(SS)基因都能够检测到的表达,但是都没有检测蔗糖磷酸合成酶(SPS)基因的表达。分析对应时期两个品种的各糖组分(包括蔗糖、葡萄糖和果糖)变化发现,奉节72-1脐橙果实的蔗糖积累在11月份就达到较高水平,葡萄糖和果糖含量在12月中旬达到最高水平,然后在果实挂树后期各糖组分表现下降趋势。而奉晚脐橙的各糖组分在果实成熟过程中一直保持上升趋势,各糖组分在元月份前的成熟过程中多数情况都显著低于对照品种的各糖组分含量。因此AI表达差异可能是造成奉晚脐橙与奉节72-1脐橙糖分差异的主要原因之一。

Expression analysis of genes of sucrose-metabolizing enzymes using RT-PCR and change of sugar contents in fruit juice sacs (edible parts) between Fengwan and Fengjie72-1 navel orange (Citrus sinensis L.Osbeck) during fruit maturation were studied in this paper. Results showed that the acid invertase (AI) gene expression in Fengwan orange was different from Fengjie72-1 orange, while sucrose synthase(SS) gene and sucrose phosphate synthase(SPS) gene had no distinct expressing difference between the two cultivars, SS could be detected and SPS could not, during fruit maturation. As the same period, sugar accumulations including sucrose, glucose and fructose in the two cultivars were exhibited difference too. Sugar accumulation in Fengjie72-1 orange presented increasing and decreasing curve, while in Fengwan orange was kept an increasing trend from Oct. to Jan. Before January, Sugar contents in Fengwan orange were obviously lower than those in Fengjie 72-1 orange. Taken together, AI expressing difference might be one of the main reasons of sugar difference between Fengwan orange and its parental line, Fengjie 72-1 orange.
