茶儿茶素-香草醛酸性试剂检测方法的研究

Study on the Detection of Catechins in Tea Plant with Vanillin-acid Reagent

儿茶素是茶叶中主要的品质和功能成分。香草醛酸性试剂比色法是检测儿茶素含量的最常见方法,但其准确性及实用性一直受到质疑。本实验利用溶剂萃取、TLC、HPLC等方法,从特异性、影响因子角度,对儿茶素-香草醛酸性试剂显色方法的特异性、准确性进行了研究。结果显示不同物质与香草醛酸性试剂显色的特异性差异较大,其中儿茶素类的显色反应的灵敏度最高,且各儿茶素的灵敏度相当;黄酮醇、二氢黄酮醇类次之;茶皂素类最低。显色试剂中的酸种类、酸浓度、显色温度、显色时间均对显色反应有影响,建议采用1%香草醛-浓盐酸试剂,在室温下反应5min,以Y (A505nm)=aX(μmol/ml)+b 公式计算茶树中儿茶素的含量,该方法简单、快速、准确。

atechins are the mainly functional and dominant flavor compounds in tea. Vanillin colorimetric method is usually used to detect catechins content, but its accuracy has been questioned. Solvent extraction, TLC and HPLC analysis were adopted to investigate the specificity and accuracy of reaction between vanillin-acid reagent and catechins. The results demonstrated that significant differences existed in the reactions of vanillin-acid reagent with different substances, among of which catechins had the highest sensitivity, lower in flavonols and flavanonols, and lowest in tea saponin. Factors influencing the reactions of vanillin-acid reagent with catechins, including the kinds of acid, acid concentration, reaction temperature and reaction time were further investigated. The results suggested that a sensitive, simple and relatively specific method to analyze catechins content should be performed by using 1% vanillin-HCl reagent and reacting for 5 minutes under room temperature. Catechins content could be calculated with the formula of Y(A505nm)= aX(μmol/ml)+b.
