星形细胞上调基因-1（AEG-1）在胰腺癌中的表达增高并促进胰腺癌的增殖

Objective：Explore biological function of proto-oncogene AEG-1 in pancreatic cancer. Methords: RNA extraction, reverse transcription and real time quantitative polymerase chain reaction were used to detect AEG-1 expression in pancreatic cancer tissues and cell lines,compared with normal pancreatic tissue and ductal epithelium cell line. Establish a stable cell line and SiRNA transfection cell model. Real time quantitative polymerase chain reaction and western blot were used to estimate the efficiency of the transfections. Additionally, MTT assay, bromodeoxyuridine labeling and immunofluorescence were used to detect cell proliferation. Flow cytometry were used to analysis cell cycle. Results: In pancreatic cancer tissue and cell lines, AEG-1 was significantly highly expressed. MTT assay and BrdUrd signalling revealed that silencing the expression of AEG-1 suppressed，while upregulation of AEG-1 promoted pancreatic cancer cells poliferation. Additionally, knockdown AEG-1 led to G1/S arrest while overexpression of AEG-1 shorten G1 phase and promote S period. Conclusions: Proto-oncogene AEG-1 was upregulated in pancreatic cancer, which significantly enhanced the proliferation of pancreatic cancer cells, suggesting a novel tumor bio-marker in early detection of pancreatic cancer. ??
