乙酸钠和丁酸钠对奶牛乳腺上皮细胞中乳脂合成的协同作用
Effect of sodium acetate and sodium butyrate on milk fat synthesis in dairy cow mammary epithelial cells
为探讨乳脂肪合成的前体乙酸钠和丁酸钠协同作用对乳腺中乳脂合成的影响,本研究通过半自动生化分析仪和荧光定量RT-PCR方法检测了不同浓度乙酸钠和丁酸钠协同添加对奶牛乳腺上皮细胞中甘油三酯合成以及乳脂合成关键酶基因表达的影响,确定了当1mmol/L丁酸钠和8mmol/L乙酸钠协同作用时,乳腺上皮细胞中甘油三酯合成和分泌最多,同时乙酰CoA羧化酶和脂肪酸合酶的基因表达水平较高,为营养调控乳脂合成提供理论依据。
In order to clarify the effect of sodium acetate and sodium butyrate on milk fat synthesis in mammary gland, semiautomatic biochemistry analyzer was used to detect the effect of sodium acetate and sodium butyrate combined on triglycerides biosynthesis and RT-PCR was used detected ACC and FAS mRNA expression. The results showed that 1mmol/L sodium butyrate and 8mmol/L sodium acetate induced the most triglycerides production in mammary epithelial cells, and ACC and FAS mRNA expression were higher as well. It provided the theory basis for controlling milk fat synthesis by nutrition.
