中药对SiHa-CTL-Treg共培养体系中CTL细胞CTLA-4、PD-1蛋白的影响

目的:通过检测CTLA-4、PD-1的蛋白表达情况,了解清毒栓是否可以抑制Treg细胞的功能来改变局部免疫微环境而达到抑制肿瘤。方法:免疫磁珠分选Treg、CTL细胞;Western-Blot法检测CTLA-4、PD-1蛋白表达。结果:CTL与Treg共培养组,CTLA-4、PD-1的蛋白表达量明显增加,β-榄香烯及清毒栓干预后CTLA-4、PD-1的蛋白表达量显著降低,且清毒栓组蛋白表达量降低的更加明显。结论:β-榄香烯、清毒栓都可以抑制Treg对CTL中CTLA-4、PD-1蛋白的上调作用。
Objective: detecting the protein expression of Cytotoxic T lymphocyte antigen-4 (CTLA-4) and Programed Death-1 (PD-1), we try to prove that Qingdu suppository could suppress Treg cell function which would change the local immune microenvironment and inhibit tumor growth. Methods: the Treg and CTL cells were separated by immunomagnetic separation. The protein expression level of CTLA-4 and PD-1 was studied by Western-Blot. Results: the expression trends of CTLA-4 and PD-1 were increased in the CTL/Treg co-colture gruop. In CTL+Treg+SiHa cell cultural supernatant+β-elemene group and CTL+Treg+SiHa cell cultural supernatant+ Qingdu suppository group, the protein expression of CTLA-4 and PD-1 decreased significantly, however, the CTL+Treg+SiHa cell cultural supernatant+ Qingdu suppository group was lower of that than the CTL+Treg+SiHa cell cultural supernatant+β-elemene group. Conclusion: both of elemene and Qingdu suppository could suppress the Treg cell up-regulation function of CTLA-4 and PD-1 protein expession in CTL cell.
