目的:研究分子量50万、脱乙酰度为90%的壳聚糖给小鼠灌胃后在体内的吸收、分布及排泄。 方法: 用异硫氰酸荧光素对壳聚糖进行标记,确定荧光标记壳聚糖在小鼠组织中的检测方法,测定小鼠口服荧光标记壳聚糖后在体内各组织中的分布,并检测给药后72h内药物从尿和粪中排泄情况。结果:获得纯化的荧光标记壳聚糖,各组织和排泄样品中低、中、高浓度的回收率、精密度和稳定性均符合方法学技术要求。给小鼠灌胃后壳聚糖在血清中浓度非常低,体内主要分布在肾脏,肝脏其次。尿液中壳聚糖被分解,粪便中壳聚糖以原形排泄。结论:壳聚糖在心、肝、脾、肺、肾和肠中均有分布,胃部没有吸收,主要经肾脏排泄。高分子量壳聚糖在小鼠体内的组织分布的研究为其进一步的开发具有指导价值。

Objective:A chitosan sample with molecular weight Mw (5×105)and the degree of deacetylation DD(>90%) was prepared to study the distribution and excretion of the Chitosan in mice after oral administion. Method: Chitosan was labeled with fluorescein isothiocyanate.,and method was identified in which Chitosan fluorescent was detected in mouse tissues. Distribution of labled chitosan in organs and excretion of it in 72 hour were detected in mices by fluorescence spectrophotometer. Results: Fluorescent labelled chitosan was purified. Organizations and excretion samples of low, middle and high concentrations of recovery, precision and stability requirements are in line with methodology. Concentration of Chitosan in serum is very low in mice oral administion.It was mainly distribute in the liver. Chitosan in urine was broken down and excreted with prototype in feces. Conclusion: Chitosan was distributed to those tested such as heart,liver,spleen,lung,kidney and intestinal and so on and there was no distribution in stomach, kidney was the main excretion way.The research is essencial to the development of chitosan in pharmaceutical and biomedical applications.
