目的 观察热休克转录因子1在高迁移率族蛋白1引起的炎症反应中的表达及作用。方法 HMGB1(0-1000ng/ml)作用于RAW264.7细胞后于指定时间收集细胞上清,酶联免疫吸附法测定TNF-α水平以观察HMGB1引起TNF-α表达的时效及量效关系。免疫荧光法检测HMGB1引起的NF-κB核转移。蛋白免疫印迹法检测HMGB1引起的HSF-1表达。应用反义RNA沉默HSF-1的表达后,检测其对HMGB1诱导的TNF-α表达的影响。结果 (1)HMGB1作用与细胞后4h, 12342.4±467.4 pg/ml及12h, 19568.3±569.2 pg/ml出现两次分泌高峰(P<0.01,F=1342.7);TNF-α的表达随HMGB1使用量的增加而增加(P<0.01,F=1023.3)。(2)HMGB1可以引起NF-κB的核转移。HMGB作用后NF-κB位于胞核中的阳性率0h: 5.2±0.2%,0.5h: 32.2±2.3%,1h:89.3±5.2%,(P<0.05,F=15.2)。(3) HMGB1可以引起HSF-1表达的增加,0.675±0.022 vs 0.430±0.030, (P<0.05, t=10.8)。(4)应用siRNA抑制HSF-1的表达后,HMGB1诱导产生更大剂量的TNF-α,14470.3±432.7pg/ml vs 19388.5±557.6pg/ml, (P<0.05, t=15.6)。结论 HMGB1作用于RAW264.6细胞后4h和12h出现两次分泌高峰;TNF-α的表达随HMGB1使用量的增加而增加;HMGB1可以引起NF-κB的核转移;HMGB1可以上调HSF-1的表达;HSF-1抑制了HMGB1诱导的TNF-α表达。

Objective To observe the expression of heat shock transcription factor 1 in inflammation induced by high mobility group protein B1 and explain its effect. Methods RAW264.7 cell were stimulated with HMGB1 (0-1000ng/ml), and the supernatant was assayed for TNF-α by ELISA at the indicated time after stimulation to observe the HMGB1 time-dependent and does-dependent stimulation of TNF-α. Translocation of NF-κB was observed by immunofluorescence. Expressions of HSF-1 in cell lysates were detected by Western blot. Cell were transfected by siRNA then stimulated with HMGB1, and the supernatant was assayed for TNF-α by ELISA. Results (1) Peaks of TNF-α were observed 4h and 12h after stimulation of HMGB1(12342.4±467.4 pg/ml, 19568.3±569.2 pg/ml, P<0.01,F=1342.7). The expression of TNF-α were increased with the increased does of HMGB1(P<0.01,F=1023.3. (2) Translocation of NF-κB was observed after stimulated with HMGB1: 0h: 5.2±0.2%,0.5h: 32.2±2.3%,1h:89.3±5.2%,(P<0.05,F=15.2). (3) upregulation of HSF-1 was observed after stimulated with HMGB1: 0.675±0.022 vs 0.430±0.030,(P<0.05, t=10.8). (4) The expression of TNF-α were increased after transfected siRNA of HSF-1, 14470.3±432.7pg/ml vs 19388.5±557.6pg/ml, (P<0.05, t=15.6). Conclusion The expression of TNF-α after stimulation of HMGB1 has two peaks 4h and 12h; The expression of TNF-α were increased with the increased-does of HMGB1; HMGB1 induced translocation of NF-κB; HMGB1 upregulated the expression of HSF-1; HSF-1 inhibited the expression of TNF-α induced by HMGB1.
