为了研究PGC-1α表达与肌纤维类型转化的关系,我们以二花脸、皮特兰猪为动物模型,检测不同肌肉组织中PGC-1α、MEF2 mRNA的表达情况以及肌纤维组成。结果显示,在背最长肌PGC-1α mRNA以及MyHCs比例在品种间没有显著的差异,二花脸MEF2 mRNA的表达显著高于皮特兰(p < 0.05)。在半腱肌,二花脸PGC-1α 、MEF2 mRNA的表达均显著高于皮特兰猪(p < 0.05),而且二花脸半腱肌MyHC II x mRNA的比例极显著低与皮特兰(p < 0.01),MyHC II b 有低于皮特兰的趋势(p = 0.093)。结果提示,猪肌肉PGC-1α – MEF2 与肌纤维类型的转化有关,但是PGC-1α不是肌纤维类型转化的决定性因素。

We detected PGC-1α (PPARγ coactivator 1 alpha) mRNA expression as well as myofiber type composition in longissimus dorsi and semitendinosus muscle in Erhualian and Pietain to reveal the relationship of PGC-1α and myofiber type composition. Results shown that in longissimus dorsi, there were no significant differences of PGC-1α mRNA expression and myofiber type composition (indicated by MyHCs mRNA percentage), although MEF2 mRNA in Erhualian was expressed significantly high than in Pietrain (p < 0.05). While in semitendinosus muscle, there was significantly higher PGC-1α, MEF2 mRNA (p < 0.05) in concert with lower MyHC II x (p < 0.05) and II b (p = 0.093) expressed in Erhualian than in Pietrain. Taken together, PGC-1α might have an important role in myofiber type tranformation, but not the crucial factor.
