Effect of the TOR protein inhibitor rapamycin against Phytophthora nicotianae
目的:由烟草疫霉(Phytophthora nicotianae)引起的烟草黑胫病是烟草主要病害之一,对烟草产业造成了极大的经济损失。在化学药剂对环境损害日益增大的情况下,环境友好的烟草黑胫病绿色防治方法需求迫切。方法:近来的研究显示TOR在真核生物的生长发育过程中具有重要作用,因此本实验以烟草疫霉TOR(PnTOR)为靶标,研究TOR蛋白的特异性抑制剂雷帕霉素(rapamycin,RAP)对烟草疫霉菌生长发育的调控。结果:首先我们在烟草疫霉基因库筛选出了烟草疫霉TOR(PnTOR)蛋白质序列,并与拟南芥、人和酵母序列比对后构建进化树,并分析了烟草疫霉中TOR 信号途径的核心组分,结果表明烟草疫霉中存在保守的TOR 信号通路。进一步使用含有不同浓度的雷帕霉素的培养基培养烟草疫霉,结果表明雷帕霉素能够显著抑制烟草疫霉的生长。结论: PnTOR基因对于烟草疫霉的生长发育有重要作用,以PnTOR为靶标防治烟草疫霉病害的发生具有潜在可行性。)
Objective: Tobacco black shank caused by Phytophthora nicotianae Breda de Haan is the main diseases of tobacco, which has caused great economic losses to the tobacco industry. In the case of increasing environmental damage to chemical agents, we are committed to studying innovative green prevention and control methods. Method: Recent studies have shown that TOR plays an important role in eukaryotes growth and development, so we conducted experiments to study the role of PnTOR in Phytophthora nicotianae growth and development. Results: Screening Phytophthora nicotianae TOR signaling components and aligning the amino acid sequence of PnTOR with Arabidopsis, human and yeast TOR showed that TOR signaling pathway is conserved evolution in Phytophthora nicotianae. To investigate the role of the TOR gene in Phytophthora nicotianae, different concentrations of the TOR protein inhibitor rapamycin (RAP) was added into the medium to culture Phytophthora nicotianae. The results showed that the growth of Phytophthora nicotianae was significantly inhibited by rapamycin. Conclusion: These data indicated that TOR gene plays an important role in the growth of Phytophthora nicotianae, which lays a foundation for deeply studying the growth and pathogenicity mechanism of TOR gene.
