逍遥散对肝郁脾虚证大鼠海马区GluR1 mRNA
Effect of XiaoYaoSan on gene expressions of GluR1 in Hippocampus of rats with liver depression and spleen defi -ciency syndrome caused by chronic immobilization stress
目的:通过检测逍遥散对慢性束缚应激致肝郁脾虚证大鼠海马CA1、CA3区GluR1基因表达,明确其作用是否与杏仁核注射AMPA受体拮抗剂CNQX后对海马起到的保护作用的机理一致。方法:将30只雄性S D大鼠随机分为正常组(A组)、假手术组(B组)、模型组(C组)、逍遥散组(D组)、CNQX组(E组)和逍遥散+CNQX组(F组)。C、D、E、F组大鼠通过连续21 d (每天束缚3 h) 慢性束缚应激建立肝郁脾虚证候模型,D、F组大鼠每天束缚前灌服逍遥散5.32 g/kg,E、F组大鼠在第1、4、7、10、13、16、19、21d右侧杏仁核区微量注射α-氨基羟甲基恶唑丙酸(AMPA)受体拮抗剂CNQX 0.5 μl。第22d腹腔深度麻醉后处死大鼠,用实时荧光定量PCR的方法检测海马CA1和CA3区GluR1 mRNA基因表达的变化。结果:慢性束缚应激致肝郁脾虚模型组大鼠GluR1在海马CA1区的表达,与正常组相比都有增加的趋势,而逍遥散组、CNQX组和逍遥散与CNQX联合组对此都有一定的下调作用;模型组GluR1在CA3区的表达,与正常组相比都有减少的趋势,而逍遥散组、CNQX组和逍遥散与CNQX联合组对此都有一定的上调作用。结论:逍遥散对肝郁脾虚证的作用,与杏仁核注射AMPA受体拮抗剂后对海马起到的保护作用机理相同。
o observe the effect of XiaoYaoSan on the gene expressions of GluR1 in hippocampus CA1and CA3 areas of rats with liver depression and spleen deficiency syndrome caused by chronic immobilization stress, to discuss whether the mechanism of XiaoYaoSan is similar with the protection to hippocampus after intra-amygdala infusion of the AMPA receptor antagonist CNQX. Methods:30 male SD rats were randomly divided into normal control group (A group), sham-operation group (B group), model group (C group), XiaoYaoSan group (D group), CNQX group (E group) and XaoYaoSan + CNQX group(F group). Rats of C, D, E, F groups were restrained 3 hours everyday for 21days to establish the model of liver depression and spleen deficiency syndrome caused by chronic immobilization stress (CIS). Xiaoyaosan was administered from the first day to the 21st day before the immobilization in D and F groups. On the 1st, 4th, 7th, 10th, 13th, 16st, 19th, 21st day, unilateral intra-amygdala infusion of the AMPA receptor antagonist CNQX (0.5μl) was administered to rats of E and F groups. Rats were executed on the 22nd day, then real-time qPCR was used to compared the gene expression of GluR1 mRNA in hippocampus CA1and CA3 areas. Results:the gene expression of GluR1 mRNA in model group in hippocampus CA1 area were increased compared with normal group, XiaoYaoSan group, CNQX group and XaoYaoSan+CNQX group could down regulate the mRNA gene expression. The mRNA gene expression of GluR1 in model group in hippocampus CA3 area were decreased compared with normal group, XiaoYaoSan group, CNQX group and XaoYaoSan+CNQX group could up regulate the mRNA gene expression . Conclusion: the mechanism of XiaoYaoSan for the treatment of liver depression and spleen deficiency syndrome may be similar with the protection to hippocampus after intra-amygdala infusion of the AMPA receptor antagonist CNQX to rats.
