微孢子虫对家蚕幼虫组织侵染的PCR检测
he Characteristics for Nosema Bombycis to Infect Part Tissues of Silkworm Larva
家蚕微孢子虫(Nosema Bombycis)引起的家蚕微粒子病能对蚕桑业造成极大的损失。为了进一步研究家蚕微孢子虫对家蚕幼虫组织的侵染特性,五龄起家蚕经口注入10μL浓度为107/ml的微孢子虫悬液,分别在接种后的6h 、12h、24h、36h、48h、72h、96h、120h、144h、168h解剖家蚕,剖取中肠、丝腺、生殖腺、马氏管、表皮组织,提取基因组DNA然后用PCR检测。结果显示各组织感染时间为中肠24h、丝腺48h、生殖腺72h、马氏管72h、表皮72h。
he pebrine caused by Nosema bombycis cause a huge damage to sericultural industry. The characteristics for N. bombycis to infecting silkworm larva was studied. Silkworm of fifth instar were injected with 10μL suspension of N. bombycis which concentration is 107/ml. The silkworm were dissected at 6h 、12h、24h、36h、48h、72h、96h、120h、144h and 168h to get midgut 、silk gland、gonad、malpighian tubule and epidermis Which genomic DNA were extracted and detected by PCR. The result were that DNA of N. bombycis could be detected in midgut, silk gland, gonad, Malpighian and epidermis respectively at 24h, 48h, 72h, 72h, 72h.
