肾发育中增殖相关蛋白的表达与凋亡细胞的分布
he expression of proteins relating to proliferation and the distrussion of apoptotic cells of renal development
目的:本研究通过传统的和改良的组织化学方法显示生后14天小鼠肾增殖相关蛋白Ki67和凋亡细胞的分布。方法:取生后14天的小鼠,经灌流固定后,取双侧肾组织,分别制备石蜡和树脂812包埋的组织块。石蜡切片5μm厚,树脂切片2.5μm厚。在石蜡切片上进行传统的免疫细胞化学染色,而在树脂切片上进行改良的免疫细胞化学染色,随后检测增殖相关蛋白Ki67和凋亡阳性细胞(Tunel染色)的表达。结果:树脂切片与石蜡切片上Tunel阳性细胞的表达一致,分布比较广泛;但Ki67只在石蜡切片制备的肾组织上特异性的表达在髓放线的小管细胞核上。结论:树脂切片上进行免疫组化虽然能在特异性的显示组织蛋白的同时展示较好的组织结构,但是其免疫组化反应仍有局限性。生后肾发育时凋亡现象较普遍,增殖相关蛋白主要表达在近端小管细胞核上。
Objective: This research demonstrated the distribution of mice kidney proliferation related proteins Ki67 and the apoptotic cells after p14d by traditional and modified organization chemical methods. Methods: After 14 days of mice’s birth, we took their bilateral renal organization after their bodys were perfusion fixed, to make epoxy and resin 812 embedding tissue mass, and make epoxy section 5μm and resin slice 2.5μm in thick. Next step, the resin slice was processed by traditional immunecytochemistry dyeing, and the epoxy section was processed by improved immunecytochemistry dyeing, afterward we detect expression of Ki67 and apoptosis positive cells. Result: The expression of apoptotic cells both on epoxy and resin section were consistent, and have more extensive distribution, while Ki67 was specifically expressed only on the kidney tissues made from resin section and only expressed on tubule nucleus processed by medullary ray. Conclusion: Although immunohistochemistry on resin slice can display in specific tissue protein and show better organization structure, but its immunohistochemical reaction still has limitations. After birth, during renal development apoptosis become common phenomenon, proliferation related proteins were mainly expressed in the proximal tubular nuclei.
