以美国黄松成熟胚为外植体诱导再生小植株试验表明,成熟胚不定芽诱导以GD培养基最好,附加1mg稬-1的6-BA时,诱导率可达60%以上;不定芽增殖系数与蔗糖浓度呈反比,在1/2GD培养基上不定芽增殖系数最大时(6.5)的蔗糖浓度为10.61g稬-1。培养基中加入适量活性炭能促进胚生长速度,抑制不定芽分化。不定根诱导以1/2GD培养基为佳,生根率随着嫩梢高度(>2.0cm)的增加而增大,当嫩稍高度>4.0cm时,生根率可达15%。在1/2GD培养基上,生根率随着NAA浓度增大而呈指数增长,适当增大NAA浓度(>1.0mg.L-1)能极大地提高不定芽生根率;当NAA浓度为1.0mg稬-1和GA3浓度为0.5mg稬-1时,美国黄松不定芽在1/2GD培养基上的生根率最高达16.7%。在离体培养条件下,以种胚为外植体获得了再生小植株。

he mature embryo of Pinus ponderosa was used as explants to induce adventitious buds and plantlet formation. The results showed that the best medium for adventitious buds inducement is GD adding with 1mg昄-1 6-BA for a 60% abduction rate. The proliferation rate of adventitious buds varies negatively with sucrose concentration, the optimal sucrose concentration is 10.61g.L-1 on 1/2GD medium with a proliferation rate 6.5. The activated carbon properly adding in medium could promote embryo growing and restrain buds differentiation. The best medium for adventitious root formation is 1/2GD, the rooting rate increases with the adventitious shoot height (>2cm) increasing, the rooting rate is 15% under the shoot height >4cm. On 1/2GD medium, the rooting rate rises exponentially as NAA increasing, the rooting rate of adventitious buds will be greatly enhanced under a relative high NAA concentration (>1.0mg.L-1). On the 1/2GD medium adding with NAA1.0mg稬-1andGA30.5mg稬-1, the rooting rate of adventitious buds is up to 16.7%. The present experiment shows that root was successfully induced from cultured embryo of Pinus ponderosa and plantlet was regenerated from adventitious buds.
