K1在肝脏疾病中的研究进展
Recent progresses of transforming growth factor-β activated kinase 1 function in liver diseases
转化生长因子β活化激酶1(Transforming growth factor-β-activated kinase 1,TAK1)是细胞内的重要信号转导分子,本质上为丝氨酸/苏氨酸蛋白激酶,功能和序列上与丝裂原活化蛋白激酶激酶激酶(Mitogen-activated protein kinase kinase kinaser,MAPKKK,MAP3K)相关,属于MAPKKKs蛋白家族成员,故又名MAP3K7或者MEKK7。TAK1生物学作用几乎涉及细胞功能的各个方面,包括细胞的增殖、分化、存活、凋亡等及炎症免疫应答。近几年研究发现,其在肝脏疾病的发生发展中也发挥了重要作用。本篇文章将主要围绕TAK1的信号通路及其在肝细胞癌、肝纤维化、脂肪肝和慢性乙型肝炎中的功能进行综述。
ITransforming growth factor-β-activated kinase 1 (TAK1) is a important adaptor protein for intracellular signaling transduction. It belongs to Serine/threonine protein kinase and has similar function and amino acid sequences as other members of Mitogen-activated protein kinase kinase kinaser (MAPKKK, MAP3K) family. It is also called MAP3K7 or MEKK7. The biological function of TAK1 has been characterized as a key regulator in cell proliferation, differentiation, survival and apoptosis, as well as inflammatory and immune signaling pathways. Recently, TAK1 was found to be involved in the development and progression of liver diseases. This review highlights the TAK1-mediated signaling pathway and physiological functions of TAK1 in hepatocellular carcinoma, hepatic fibrosis, hepatosteatosis, and chronic hepatitis B.
