磁选育浸矿菌种新方法的研究--磁黄铁矿选育菌种初探
New Methods on Screen Strain by Use the Principle of Magnetic Separation
硫化矿浸矿细菌主要有三大种属,如何筛选这些细菌并进行遗传改良是提高金属浸出速率的关键。以矿物基质做培养基驯化、选育浸矿细菌是一种选育高效浸矿菌种常用的手段。磁黄铁矿因其含有Fe和S元素而常被用作矿物培养基,其晶格中的部分铁原子往往会被Ni,Co,Mn和Cu原子所替代,使其Fe/S不同,磁性也会发生一系列的变化。因此,可以用磁铁分选出具有不同磁性的磁黄铁矿作为矿物培养基,驯化、选育出针对不同磁性组成磁黄铁矿的浸矿细菌。本文主要分析和研究了浸矿细菌在不同磁性的磁黄铁矿培养基中的生长情况,驯化选育了原始混合菌种,得到了几株亚铁氧化活性较高的菌株。实验结果证明,采用不同磁性的磁黄铁矿驯化、选育浸矿细菌是一种快速可行的方法。
here are three main species of the sulfide bioleaching bacteria. The key of raising the bio-leaching rate is how to screen the strain and improve the genetic specificity. It is a common method to screen great efficient bioleaching bacteria that to use minerals as culture medium. Pyrrhotite is usually used as mineral culture medium for including Fe and S element. The replacement of Fe atom by Ni, Co, Mn and Cu atom in the pyrrhotite space lattice made the Fe/S and magnetic different. Therefore, the different magnetic pyrrhotite can be separated by magnet and used as mineral culture to screen bioleaching bacteria, which is the best bacteria to bioleaching the different magnetic pyrrhotite. The growth condition of bacteria is analyzed and researched in the different magnetic pyrrhotite culture, domesticated the origin bacteria and gained several active bacteria with fast oxidation of Fe2+.
