二酰甘油酰基转移酶 ( Acyl CoA:diacylglycerol acyltransferase, DGAT) 催化三酰甘油(TAG)生物合成最后一步酯酰化反应,是决定碳源流向TAG的一个关键酶,在TAG合成和积累中起重要作用。旨在对富集GLA的产油真菌卷枝毛霉(Mucor circinelloides)DGAT基因进行初步探索,利用RT-PCR克隆卷枝毛霉DGAT基因全长cDNA序列,测序验证,并对DGAT蛋白质序列进行生物信息学分析。结果表明卷枝毛霉存在4个DGAT基因,分别命名为McDGAT1、McDGAT2、McDGAT3和McDGAT4,依次对应JGI编号154800/154801、167165、144132和187167,其中,JGI登录编号154800与154801共同编码一个完整DGAT蛋白质;McDGAT1、McDGAT4属于DGAT2 亚家族,McDGAT2、McDGAT3属于DGAT1 亚家族,并且4个DGAT蛋白质具有不同的理化性质、二级结构和跨膜区域。研究结果为进一步研究卷枝毛霉DGAT基因结构与功能提供了重要信息。

cyl-CoA:diacylglycerol acyltransferase (DGAT) catalyzes the final step in the TAG biosynthetic pathway esterifying DAG to TAG. DGAT plays a key role in determining the carbon flux into TAG. To understand structure/function of DGATs in Mucor circinelloides, we cloned the DGAT gene and analyzed the bioinformatics. The results showed that 4 distinct DGAT genes were identified in Mucor circinelloides, designated McDGAT1, McDGAT2, McDGAT3 and McDGAT4, respectively. McDGAT2 and McDGAT3 into the DGAT1 clade, and McDGAT1 and McDGAT4 grouped into the DGAT2 subfamily.The four putative DGATs were different from in Biochemical properties, secondary structure and transmembrane helices. This study would provide important information for further studies of DGAT gene structure and function in Mucor circinelloides.
