M3受体与β-catenin在大鼠心室肌结构共定位关系的研究

目的 研究M3受体与粘连蛋白β-catenin在大鼠心肌缺血时的结构相互作用,为筛选抗心律失常药物新靶点提供依据。方法 结扎大鼠左旋冠状动脉前降支建立心肌缺血模型,取心脏组织,并分别用激光共聚焦显微镜及免疫沉淀与免疫印迹方法,研究M3受体与β-catenin在大鼠心室肌结构共定位关系。结果 在大鼠心肌细胞膜上M3与β-catenin存在结构共定位关系,免疫沉淀发现随着缺血时间的延长,共定位关系加强;在急性心肌缺血模型中胆碱能增强两者的结构共定位关系。结论M3与β-catenin在大鼠心室肌细胞存在结构共定位关系,这种共定位关系可能是新的抗心律失常药的作用靶点。

im To study structural integration between M3 muscarinic acetylcholine receptors (M3-mAChR) and β-catenin in parts of cell membranes in isolated myocytes and explore the new target drug action for antiarrhythmia.Methods Immunostaining the single normal ventricular myocytes were introduced to determin the distribution of M3-mAChR and β-catenin. Immunoprecipitation was used to demonstrate that the colocalization between M3-mAChR and β-catenin in response to the ischemia time developing. Results β-catenin was closely associated with M3-mAChR in parts of cell membranes in isolated Myocytes. At the same time, the colocalization was closer in response to the ischemia time developing and choline partially strengthened the ischemia-induced activation of the association between β-catenin and M3-mAChR. Conclusion The structural integration between M3-mAChR and β-catenin in rat ventricular myocardium,and this integration could be strengthened by administration with choline.
