维生素D联合n-3多不饱和脂肪酸促进MCF-7细胞凋亡

乳腺癌是女性中的主导型癌症。n-3多不饱和脂肪酸(n-3 PUFAs)和维生素D在降低乳腺癌发病率中都有着积极作用。然而, n-3 PUFAs联合维生素D是否在乳腺致癌中具有更强的保护作用,这一点仍不清楚。在这项研究中,我们先用不同浓度的n-3 PUFAs和VD3分别处理MCF-7细胞,再用显微镜观察和血球计数法检测两种物质单独对细胞生长的影响,之后将两种物质叠加观察对细胞生长的影响,用DAPI染色观察对细胞凋亡的作用,并用免疫杂交法分析MAPK通路在其中的作用。本文发现,n-3 PUFAs和1α,25-二羟基-维他命D3(VD3)联合处理后,显著增加了MCF-7细胞凋亡的产生,上调了MAPK通路中Erk1/2和p38的磷酸化水平,并且Shp2也部分参与了联合所诱导的细胞凋亡。因此,维生素D和n-3多不饱和脂肪酸的联合通过激活MAPK途径诱导MCF-7细胞的凋亡。
Breast cancer is a leading type of cancer in women. Both omega-3 polyunsaturated fatty acids (n-3 PUFAs) and vitamin D play positive role in the reduction of breast cancer incidence. However, whether combination of n-3 PUFAs and vitamin D has stronger protective effect on breast carcinogenes is still remained elusive. In this study, we treated MCF-7 cells with different dose of n-3 PUFA or VD3 or both, and then cell growth was examined by cell morphology and viable cell counting assay, cell apoptosis was observed by DAPI staining, and then the effect of MAPK pathway was determined by immunoblotting. Results showed that combination of n-3 PUFAs and vitamin D enhanced cell apoptosis and increased the phosphorylation level of Erk1/2 and p38, and Shp2 played an important role in the apoptosis. To conclude, combination of n-3 PUFAs and vitamin D induces cell apoptosis in MCF-7 cells via activating MAPK pathway.
