Study on the Enhancement of Cytotoxic Effects of Gemcitabine in Combination with Elemene on Pancreatic Cancer Cells

目的:研究榄香烯对胰腺癌Bxpc-3细胞株吉西他滨化疗敏感性的影响。方法: MTT法检测不同浓度榄香烯与吉西他滨联用或单处理后胰腺癌细胞Bxpc-3的增殖率;采用等效线图解法观察联合用药是否具有协同效应;Annexin V/PI双染色法分析细胞凋亡的变化。结果:与吉西他滨单独处理48hr(IC50=0.31±0.04 μg/mL)相比,榄香烯与吉西他滨联合处理可显著降低吉西他滨的IC50(IC50=0.063±0.01μg/mL),且联合用药具有协同效应。流式细胞技术也证实榄香烯协同吉西他滨对人胰腺癌Bxpc-3细胞的早期凋亡显示出明显的时间和剂量依赖关系。结论:榄香烯与吉西他滨联合用药对人胰腺癌Bxpc-3细胞可产生协同杀伤作用,并可增强吉西他滨诱导肿瘤细胞凋亡的作用。

Objective: To explore the efficacy of elemene in combination with gemcitabine on pancreatic cancer Bxpc-3 cells. Methods: The growth inhibitory effects of elemene and gemcitabine alone or combination on Bxpc-3 cell line were assayed with MTT test. The equivalent method line method was employed to observe whether combination shown synergistic effect. Apoptosis rates were quantitatively determined by AnnexinV/PI double staining. Results: elemene and gemcitabine combination treatment on Bxpc-3 cell line for 48hr(IC50=0.063±0.01μg/mL)could reduce the IC50 of gemcitabine treatment alone for 48hr(IC50=0.31±0.04 μg/mL)significantly. Meanwhile the combination treatment shown synergistic effect. The occurrence of apoptosis was detected with a dosage-dependent and time-dependent manner by AnnexinV/PI double staining,. Conclusion: elemene in combination with Gemcitabine can effectively inhibit the growth and induce cells apoptosis in Bxpc-3 cells.
