乳牛乳房炎无乳链球菌内蒙古分离株SIP基因克隆与序列分析

根据Genbank 公布的牛源无乳链球菌SIP基因序列,设计并合成一对引物,通过PCR 扩增,获得SIP基因部分扩增产物。序列分析结果表明,SIP基因扩增产物大小为945bp, 负责编码315个氨基酸残基。标准菌株(+株)与内蒙古分离株(M株)的基因及氨基酸同源性100%,该两个菌株与Genbank上公布的B群无乳链球菌菌株(DQ914274)SIP基因及氨基酸同源性达到98.94%及99.68%。高度同源性结果为进一步研究和开发新型有效免疫制剂及诊断试剂奠定了理论和实验依据。
pair of primers were designed and synthesized according to SIP gene sequences of bovine mastitis Streptococcus agalactiae published on Genbank. SIP gene fragment were amplified by PCR。Sequencing result showed that SIP gene was 945bp in length, which were encoding 315 amino acid residues. The standard strain (+strain) and isolated strain from Inner Mongolia (M strain) had 100% of homology in genes and amino acid. The SIP Gene and amino acid sequence of the two strains and published Streptococcus agalactiae group B strain (DQ914274) on genbank were homology reach 98.94% and 99.68%, respectively. The result of a high degree of homology has laid a theoretical and experimental basis for further research and development of new effective immune agents and diagnostic reagents.
