SKIP调控SEF pre-mRNA的可变剪接抑制拟南芥开花时间的分子机制
SKIP regulates flowering time through alternative splicing of SEF pre-mRNA in Arabidopsis
在酵母和哺乳动物中,Ski-Interacting Protein (SKIP) 作为剪接因子和转录的共激活或共抑制因子,调节基因的表达。SKIP参与生物钟相关基因及盐胁迫相关基因前体mRNA(pre-mRNA)的可变剪接,调控拟南芥生物钟和耐受盐胁迫的能力。此外,SKIP还参与开花时间的调控,但是分子机制尚不清楚。本研究发现,SKIP是开花抑制因子,通过调控SWR1染色质重塑复合体的组分SEF pre-mRNA的可变剪接,促进开花抑制因子FLC的表达,抑制开花促进因子SOC1、FT、TSF的表达,延迟拟南芥开花。本研究揭示了SKIP调控开花时间分子机制,为深入研究SKIP调控开花时间的生化机制奠定了基础。
In yeast and mammals, Ski-Interacting Protein (SKIP) plays important roles as a transcriptional co-regulator and splicing factor in regulating gene expression. SKIP is required for manipulating the function of circadian clock and tolerance to salt stress through alternative splicing in Arabidopsis. In addition, SKIP is involved in flowering time control. However, the molecular mechanisms remain obscure. In this study, the roles of SKIP in controlling flowering time are investigated. Our results show that SKIP is a suppressor of flowering time. It promoter expression of FLC through alternative splicing of SEF pre-mRNA, a component in chromatin remodeling SWR1-complex, represses the expression of SOC1, FT, and TSF, delays flowering time in Arabidopsis. This study reveals the molecular mechanisms of SKIP on controlling flowering time, which lays a sound foundation for further investigating the biochemical mechanisms.?????
