复方银黄微型灌肠剂对大鼠腹腔巨噬细胞NO及iNOS的影响
Effect of Yinhuang Micro-enema Compound on nitric oxide and induced nitricoxide synthase in rat Peritoneal macrophage
目的:研究复方银黄微型灌肠剂(Yinhuang Micro-enema Compoumd,YHMCE)对巨噬细胞释放NO及iNOS(诱导型一氧化氮合成酶)活性的影响,探讨其抗炎作用分子机理。方法:体外培养大鼠腹腔巨噬细胞(Peritoneal macrophage,PMФ),采用氧化还原酶法、免疫细胞化学法和RT-PCR分别测定不同剂量YHMCE对LPS诱导大鼠腹腔巨噬细胞NO生成量、iNOS活性及iNOS mRNA表达水平。结果:YHMCE能降低大鼠腹腔巨噬细胞iNOS活性及iNOS mRNA表达水平,减少LPS诱导的NO大量产生和释放,并呈作用剂量依赖关系。结论:YHMCE对大鼠腹腔巨噬细胞NO产生的调节可发生在iNOS的转录水平,抑制iNOS 从头合成,减少NO合成与释放,可能是其发挥抗炎作用的分子作用机理之一。
IM To study the effect of Yinhuang Micro-enema Compound(YHMCE) on release of nitric oxide(NO) and the activity of induced nitricoxide synthase(iNOS) so as to elucidate its anti-inflammatory mechanisms. METHODS Peritoneal macrophage(PMФ) obtained from normal rats were cultivated in vitro. The production of NO,the activity and mRNA expression of iNOS in peritoneal macrophages of mice administered with different dose of YHMCE were observed by nitrate reductase method,immunocytochemical method and RT-PCR,respectively. RESULT YHMCE can inhibit the activity and mRNA expression of iNOS with dose-dependence in rat peritoneal macrophages, and reduced the generation and release of NO. CONCLUSION The regulation of YHMCE on the production of NO in peritoneal macrophages of mice may occur at transcriptional level of iNOS, inhibiting the iNOS’s denovo synthesis and reducing the the generation and release of NO .This may be one of its anti-inflammatory mechanisms.
