尼罗罗非鱼Sox9a基因的敲除研究

Sox9在脊椎动物中参与性别决定与分化及性腺发育。本研究对尼罗罗非鱼中哺乳类Sox9基因的同源体Sox9a在生殖发育中的功能进行了初探。利用TALEN基因敲除技术对尼罗罗非鱼Sox9a基因进行敲除发现,Sox9a敲除的F0代突变雄鱼精巢中的精母细胞数量减少,精小叶退化;部分Sox9a杂合突变雌鱼卵巢中的生殖细胞缺失。这些结果表明,Sox9a在尼罗罗非鱼性腺发育,特别是生殖细胞维持中具有重要调控作用。
Transcription factor Sox9 is involved in sex determination and differentiation as well as gonadal development in vertebrate. To determine the roles of the tilapia Sox9a gene, a homolog of mammalian Sox9 gene, in reproductive development, the transcription activator-like effector nucleases (TALENs)-mediated knockout of the tilapia Sox9a gene was performed. As a result, Sox9a mutation resulted in a reduction of the number of spermatocytes and the degeneration of seminiferous tubules in the testis of F0 generation tilapia. In addition, it is noted that germ cells were lost in the ovary of several tilapia fishes with heterozygous mutation of the Sox9a gene. Taken together, these results indicate that Sox9a plays key roles in the gonadal development and maintenance of germ cells in the tilapia.
