本实验主要通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)的手段研究了豆渣蛋白(OP)成分的特性。研究表明,与大豆分离蛋白(SPI)相比,豆渣蛋白含有3种特征性蛋白条带,分子量分别约为27.21,16.00和23.38kDa。基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)的测定结果表明,分子量为27.21和23.38kDa的蛋白质为碱性7s球蛋白(Bg7s),而分子量为23.38kDa的蛋白质为油体蛋白B。豆渣蛋白的非还原、还原电泳及对角线电泳结果显示,分子量为50.10和34.27kDa的豆渣蛋白含有分子间二硫键,MALDI-TOF-MS鉴定其分别为11s球蛋白和Bg7s。豆渣蛋白的三大组分为-残留的贮藏蛋白(7s和11s),Bg7s和油体蛋白B,其相对浓度分别约为57%,31%,12%。

his experiment mainly studies the compositional characterization of okara proteins (OP)by SDS-PAGE. The results show that, when compared with Soy Protein Isolates(SPI),okara protein contains three characteristic protein bands, whose molecule weight(MW) are respectively 27.21,16.00 and 23.38kDa. By MALIDI-TOF-MS, the 27.21 and 16.00kDa protein were identified as subunits of basic 7s globulin(Bg7s), while the 23.38kDa protein were identified as oleosin isoform B. Research results of reducing, non-reducing electrophoresis and diagonal electrophoresis show that there are two proteins which are linked by subunits through disulfide bond, and they are identified as Bg7s and 11s globulin. The comparative concentration of remaining storage proteins(7s and 11s),Bg7s and oleosin was determined to be about 57%,31%,12% respectively.
