P敏感的钾通道开放剂对离体肺保护的试验研究
Protective effect of ATP regulated potassium channel opener on isolated lung
目的 研究ATP敏感的钾离子通道开放剂(PCOs)吡那地尔(Pinacidil)在离体兔肺保护中的作用。方法 将18只杂种家兔(灰色)随机分成3组,建立离体肺缺血再灌注模型。对照组(n=6)自体温血直接再灌注2小时;缺血组(n=6)用LPD(低钾右旋糖酐保存液)+DMSO(二甲基亚砜)灌洗, 4℃保存8小时后自体温血再灌注2小时;试验组(n=6)用LPD+100μmol/L吡那地尔(溶于DMSO中)灌洗,4℃保存8小时后自体温血再灌注2小时。每隔30分钟分别经肺动脉和左房插管取血行血气分析,评估离体肺功能;测量湿/干比重,估计肺水含量;光镜下行肺组织形态学检查;制做肺组织匀浆测定超氧物歧化酶(SOD)、肿瘤坏死因子-α(TNF-α)和白介素-8(IL-8)含量。结果 试验组氧分压差(ΔPo2)和SOD比缺血组明显升高(P<0.05);组织形态学检查,试验组损伤轻于缺血组;试验组的TNF-α和IL-8含量明显低于缺血组(P<0.05)。结论 在保存时间不超过8小时情况下,用加入吡那地尔的LPD液灌洗和保存肺,能更好的保护肺功能。
Objective:To investigate the protective effect of pinacidil, an ATP regulated potassium channel opener on isolated lung. Methods: 15 hybrid gray rabbits were devided into 3 groups randomly. An isolated recirculating blood perfused ventilated rabbit model were used.Control group(n=6) underwent immediate reperfused for 2 hours.Ischemia group(n=6) were flushed with LPD solution + DMSO,stored at 4℃ for 8 hours and then reperfused for 2 hours.Pinacidil protected group (n=6)were flushed with LPD+ pinacidil 100μmol/L ( soluted in DMSO) , stored at 4℃ for 8 hours and then reperfused for 2 hours. Every 30 minutes, intermittent blood gases were taken from the pulmonary artery and left atrium to evaluate the function of lungs. The histologic injury of lung samples ,the wet-to-dry ratios and the content of SOD, TNF-α,IL-8 in the homogenate of lung were measured. Results: The ΔPo2 and SOD in pinacidil group was significantly higher than in control group(P<0.05). The histologic injury in pinacidil group was less than in control group. The concentration of TNF-αand IL-8 in pinacidil group was significantly lower than in control group(P<0.05). Conclusions: The protective effects of Pinacidil in conjunctions with LPD solution on isolated lung were better during the 8 hours preservation.
