Slit2对成纤维肉瘤血管新生的影响
Effect of slit2 on fibrosarcoma angiogenesis
神经导向因子在调节神经元前体细胞定向迁移、定位和神经元轴突的定向生长中发挥重要作用。近年来发现神经导向因子也参与血管发育的调控,但对于其作用还存在很大争议。本研究比较了目前已被证实的,在血管发育过程起调控作用的几种神经导向因子在肿瘤细胞中的表达。发现slit2在人成纤维肉瘤HT1080细胞中有较高表达,而在肺癌A549细胞中不表达。为进一步检测Slit2对于HT1080诱导的血管形成的影响,使用正常人胎儿肺成纤维细胞(HFL-1)与人脐静脉内皮细胞(HUVEC)共培养诱导血管形成的模型,研究了抗体封闭Slit2后,HT1080细胞条件培养基(Condition meidum)对于血管形成的影响,发现抗体封闭Slit2可显著促进血管形成,从而证实slit2在HT1080细胞中对血管形成起负调控的作用。
xon guidance molecules play an important role in axonal growth, migration and orientation. In recent years,axon guidance molecules are also involved in the regulation of vascular growth, but with much controversy. In this study, using RT-PCR, the expression of several axon guidance molecules that had been confirmed previously playing regulatory roles in vascular development are examined in tumor cells. We show that fibrosarcoma HT1080 cells maintain a relative higher expression of slit2, whereas no expression is detected in lung cancer cells, A549. In order to further examine the effect of Slit2 on HT1080 induced angiogenesis, co-culture of normal fetal lung fibroblasts (HFL-1) and human umbilical vein endothelial cells (HUVEC) is used to induce angiogenesis, in which HT1080 cell conditioned medium acts as pro-angiogenic activator. The blockage of slit2 by antibody promotes HT1080 cell conditioned medium induced angiogenesis, thus suggesting a negative control of Slit2 in fibrosarcoma induced angiogenesis.
