兔退变腰椎间盘髓核细胞中衰老信号p16INK4a/pRb表达的体外研究

目的 分析衰老信号通路p16INK4a/pRb与椎间盘退变的关系及其表达的时效性。方法 取成年大白兔16只。实验组(L3/4、L4/5椎间盘)建立兔腰椎间盘退变模型,对照组(L2/3椎间盘)不予任何处理,术后随机分为4个模型组(术后2、4、8、12周组),每组4只,各组于相应时间点行影像学检查(MRI)和评价,随后取出髓核细胞进行体外培养,运用RT-PCR法检测各样本髓核细胞MMP3、MMP13、p16INK4a、pRb的基因表达情况,并进行定量分析。 结果 1.实验组椎间盘的髓核MRI T2WI信号强度明显降低,改良的Thompson分级随建模时间延长而逐渐加深,各时间点差异有显著性意义(P<0.05);2.随着建模时间延长,实验组椎间盘内髓核细胞 MMP3、MMP13、p16INK4a的mRNA表达量增加,pRb的mRNA表达量逐渐下降,其中在第4-8周阶段变化最为明显,且在第4、8、12周时间点比较差异有显著性意义(P<0.01)。 结论 1.随着造模时间延长,椎间盘退变程度逐渐加重,且在术后第4-8周加重趋势较强;2.随着椎间盘退变程度加重,髓核细胞中衰老信号p16INK4a的表达明显增加,pRb表达与之呈负相关。
Objective To analysis the relationship between senescence signal p16INK4a/pRb and intervertebral disc degeneration, and to makesure the expression timeliness of the signal. Methods 16 adult rabbits, the L3/4 and L4/5 lumbar intervertebral discs of each rabbit were included in the experimental group, being operated to establish degenerative disc models;the L2/3 discs were included in the control group, without any treatment. Animals were then randomly divided into four model groups(2、4、8、12-weeks post surgery), each with four. Each group were taken tested by MRI and assessed at the corresponding time point. After that, the rabbits were killed respectively.Took the nucleus pulposus cells of each sample separately, cultured under the same environment, then the gene expression levels of MMP3, MMP13, p16INK4a, pRb in each sample were detected and analyzed by RT-PCR. Results 1. The MRI signals(T2WI)intensity in the nucleus pulposus of the experimental group significantly weakened with the extension of the time point,and the improved Thompson classification of MRI gradually biased seriously, the difference in each point of time was statistically significant(P<0.05);2. In the experimental group, The mRNA expression of MMP3, MMP13, p16INK4a increased with the extension of the time point, while the mRNA expression of pRb gradually decreased. These changes were most obvious in the period of 4-8 weeks. The differences between experimental group and control group were statistically significant in the time of 4、8 and 12-weeks post surgery(P<0.01). Conclusions 1. The disc degeneration gradually became more serious with the extension of the time post surgery, and the trend was strongest in the period of 8-12 weeks post surgery. 2. As the disc degeneration became more serious, the expression of the senescence signal p16INK4a in the nucleus pulposus cells significantly increased, which had a negative correlation with the expression of pRb.
