BAMBI在C2C12成肌分化中的作用
Role of BAMBI during myogenic differentiation of C2C12
BAMBI(BMP and Activin receptor Membrane Bound Inhibitor,骨形成蛋白和激活素的跨膜抑制剂)既可以作为伪受体抑制TGFβ信号通路的活性,又可以促进经典Wnt/β-catenin通路的活性,广泛参与了机体的许多生命活动,但它对成肌分化的影响目前尚未有报道。本实验发现,BAMBI在C2C12细胞成肌分化的早期表达急剧升高,随后逐渐下降。实时定量PCR和western-blot表明,干扰BAMBI,MyoD、MyoG 和 MyHC的表达显著下降。同时,细胞免疫化学显示肌管数量、分化指数和融合指数也显著下降,提示成肌分化受到抑制。这些结果表明BAMBI可能促进C2C12成肌分化,但具体的作用机制有待进一步研究。
BAMBI (BMP and Activin receptor Membrane Bound Inhibitor) is widely involved in numerous biological processes, either through inhibiting TGFβ activity as a decoy receptor or enhancing canonical Wnt/β-catenin pathway. While its role during myogenesis remains largely unknown. In our study, BAMBI was sharply increased on the onset of myogenic differentiation, and then gradually declined. BAMBI siRNA powerfully reduced the expression level of myogenic markers, such as MyoD、MyoG and MyHC, indicated by real-time PCR and western-blot. Accordingly, cell immunochemistry assay shown a decreased number of fused myotubes, differentiation index and fusion index, which was a phenomenon of a repressed myogenic differentiation. Our data suggested BAMBI might promote C2C12 differentiation, and the underlying mechanism needs more inspection.
