血红蛋白肽分子量分布对血红素酸溶性的影响及其相互作用研究
Influence of molecular weight distribution of hemoglobin peptide on heme acid solubility and the interaction
血红蛋白中的血红素是优秀的铁源,但其单体在胃酸环境下不溶,采用酶解血红蛋白并控制分子量分布的方式可改善血红素的酸溶性。本文采用两步酶解与超滤的方法获得了不同分子量分布的血红蛋白肽,重点研究了超滤条件、血红蛋白肽分子量分布与血红素酸溶性间的联系,并通过荧光光谱探究了血红素与肽间的相互作用类型、结合常数及结合力。结果表明,超滤富集目标肽段时轻度酶解物中蛋白浓度为22.5mg/ml,轻度酶解物超滤透过液中蛋白质量与深度酶解物中血红素质量比为=1.15时产品中总血红素的酸溶性最佳,达到84.1%,小于1kDa的肽段削弱了血红素的酸溶性,而1k-2kDa的肽段则可能是提高酸溶性的关键肽段。荧光光谱法显示,血红素与肽的猝灭类型为静态猝灭,两者的主要相互作用可能以疏水相互作用为主,配位键、范德华力和氢键等引起焓变的作用力则起辅助作用。
Heme in hemoglobin is an excellent source of iron while its monomer is insoluble in acid environment of stomach. The acid solubility of heme could be improved by hydrolysing hemoglobin and controlling corresponding molecular weight distribution. In this study, hemoglobin peptides with different molecular weight distributions were obtained by two-step enzymatic hydrolysis and ultrafiltration method. The relations between ultrafiltration conditions, peptide molecular weight distributions and acid solubility of heme were investigated. Furthermore, the interaction force, binding constant and bond force between peptide and heme were explored by fluorescence spectroscopy. The results showed that acid-soluble percentages of heme achieved 84.1% when the protein concentration in light-degree hydrolysates during ultrafiltration is 22.5mg/ml and the mass ratios of protein in ultrafiltration liquid to heme in high-degree hydrolysates was 1.15. Peptide with the molecular weight is less than 1kDa weakened the heme acid solubility, whereas peptide with the molecular weight in the range of 1k-2kDa may be the key to improve the acid solubility of heme. Fluorescence spectroscopy showed that the quenching type between heme and peptide was static quenching, the main interaction force is possibly hydrophobic interactions, forces which can cause enthalpy change such as coordination bond, Van der Waals forces and hydrogen bonding play an supporting role.
