不同雌激素对大鼠颏舌肌细胞雌激素受体表达的影响比较
Effects of Different Estrogens on Estrogen Receptors Expression in Genioglossus Muscle Cell of Rats
目的:探讨不同雌激素对低氧大鼠颏舌肌雌激素受体表达的影响。方法:体外培养新生大鼠颏舌肌成肌细胞,置于含1% O2低氧培养箱内培养24 h,同时在培养基中加入不同雌激素干预,雌二醇(E2)、金雀异黄素(GEN)、白藜芦醇(RES)、白藜芦醇二聚体(RD)(浓度10-6 mol/L),另设两个对照组:空白对照组(NC)(常氧培养箱内培养)和低氧对照组(CIH)。采用Western-blot和RT-PCR法检测各组成肌细胞中ERα和ERβ蛋白及mRNA水平的表达情况。结果:各组颏舌肌成肌细胞中ERα、ERβ的核酸与蛋白水平的表达情况基本一致。ERα的表达情况:E2组表达显著升高,且明显高于其他各组(P< 0.01),GEN组、RES组、RD组显著高于CIH组,但也显著低于NC组(P<0.05),而GEN组、RES组、RD组之间ERα表达未见明显差异(P >0.05)。ERβ的表达情况:E2组表达量仍显著高于其他各组(P < 0.01),GEN组、RES组、RD组显著高于NC组(P<0.05),且该三组间存在显著差异,其中RD组>GEN组>RES组(P <0.05)。结论:金雀异黄素、白藜芦醇、白藜芦醇二聚体在ERs的选择性亲和性上主要是以ERβ为主。
OBJECTIVES:To investigate and compare the effects of different estrogens on expression of ERs in genioglossus muscle cell. METHODS:the genioglossus muscle tissues were obtained from newborn (0-3d) SD rats. The myoblasts were cultured in incubator containing 1% O2 (hypoxia condition) for 24h, and estrogens were added into the culture medium simultaneously. There were also another two control groups: the blank control group (cultured in normoxia incubator) and the hypoxia control group. Western-blotting was performed to detect the expression of ERα and ERβ at protein level in genioglossus myoblasts of rats. Real-time RT-PCR was performed to detect the expression of ERα and ERβ at mRNA level in genioglossus myoblasts of rats. Results: The results of western-blot were generally accordance with those of real-time RT-PCR. As to the expression of ERα, it was significantly higher in E2 group, compared with the other groups (P<0.01). The expression in GEN group, RES group and RD group were observed to be higher than that of CIH group (P<0.05) and lower compared with NC group (P<0.01). But no statistically significant differences were observed among the GEN group, RES group and RD group (P>0.05). The expression of ERβ of the E2 group was still the highest (P<0.01). There was a significant increase of ERβ expression in GEN, RES and RD groups in comparison with the NC group (P<0.05), and the ERβ expression was higher in the RD group than that in the GEN group and RES group (P<0.05).Conclusion: genisten, resveratrol, and resveratrol dimer exhibit higher binding affinity for ERβ than for ERα.
